Serum-free medium has become a mainstay of the medical research/biotechnology industry. In the medical research arena, serum-free media has found its place in the expansion
and differentiation of numerous cell types including adult and embroyic stem cells useful for not only replacing damaged tissue but for gene therapy studies. In the boitechnology
industry serum-free media has been used extensively for the purification of cellular secreted proteins as monoclonal antibodies and recombinant proteins.
At Quality Biological, Inc., our scientists
have been busy developing serum-free medium to facilitate your research. One of our latest formulations is a clinical grade serum-free medium, QBSF-60, conatining human pasteurized and recombinant proteins developed especially for human cord stem cell (CD34+ cells)
expansion and differentiation. Exciting data from our laboratories has illustrated the ability to ex vivo long-term engrafting capacity (see below). In addition, QBI offers
a serum-free formulation, QBSF-55, developed specifically for the growth of hybridomas including those derived using the NS-1 and Sp2/0-Ag14 myeloma fusion partners; an enriched serum-free medium,
QBSF-56 developed especially for cells that are difficult to culture; and a serum-free medium, QBSF-58, developed for murine hematopoietic stem cells.
Our Stem Cell Media: QBSF-60 is a proprietary serum-free medium designed to support human hematopoietic stem/progenitor cells (e.g., CD34+).
Our research demonstrates that the QBSF-60 formulation is a serum-free medium for CD34+ cells derived from normal bone marrow, cord blood, and peripheral blood stem cells mobilized with cytokines.
QBSF-60 Serum-Free medium is comprised of human recombinant and pasteurized proteins in Iscove's Modified Dulbecco's Medium. It has been formulated as a broad brand medium that will support the proliferation and differentiation of stem cells in the presence of the appropriate cytokines/growth factors.
Thus, QBSF-60 Serum-Free Medium has supported:
1) The ex vivo expansion of the hematopoietic stem cells (CD34+ cells) maintaining their long-term engrafting capacity as determined in both the human-NOD/SCID mouse and human-fetal sheep models.
2) The ex vivo differentiation of human hematopoietic stem cells derived from umblical cord blood, adult bone marrow, and mobilized periperal blood to the myeloid, erythroid, and lymphoid lineages.
3) The ex vivo differentiation of the hematopoietic stem cells to functional natural killer and dendritic cells.
4) The ex vivo expansion of human endothelial cells derived from umblical cord veins and coronary arteries.
QBSF-58 is a proprietary serum-free medium designed for the expansion of murine and human bone marrow cells in liquid culture. The expansion of bone marrow cells is a technique with important research and clinical benefits. Research is underway to explore the possibility of removing bone marrow from a donor and placing it in culture. The aim is to expand the various elements so that they can be used for transplantation into patients whose own marrow has been damaged by disease or therapy (radiation, chemotherapy, etc.)
Using QBSF Murine bone marrow cells, cultured for 21 days in QBSF-58 medium in the presence of the cytokines rmSCF and rmGM-CSF, increased the total cell number over 90-fold. The presence of either of these cytokines alone slightly increased the cell number from the original seeding level. In the absence of cytokines the viable cell number decreased to less than 50% of the initial seeding level.
Optimal survival enhancement and hematologic reconstitution can occur in lethally irradiated mice transplanted with cells cultured for 7-14 days in QBSF-58 medium, containing rmSCF alone or cells cultured for 7 days in QBSF-58 medium, containing both rmSCF and rmGM-CSF. Hybridization analyses of bone marrow, spleen and thymus DNA from irradiated mice transplanted with these cultured cells, confirmed donor cell derived repopulation at 6 months post-transplant.
Thus, QBSF-58 medium can support, in the presence of the appropriate cytokines, both short-term and lon-term marrow repopulating cells. QBSF-58 medium is comprised of Iscove's Modified Dulbecco's Medium supplemented with BSA, insulin, transferrin and cholesterol.
