Ham’s F-12 without Hypoxanthine and L-Glutamine
Description
Compared to other basal media, Ham’s F-12 contains a wider variety of components, including zinc, putrescine, hypoxanthine, and thymidine. Ham’s F-12 contains no proteins or growth factors, and uses a sodium bicarbonate buffer system (1.176 g/L); therefore requiring a 5–10% CO2 environment to maintain physiological pH.

Applications
Ham’s F-12 Nutrient Mixture without Hypoxanthine is useful for serum-free clonal growth of Chinese Hamster Ovary (CHO) cells, chick embryos, and mouse L-cells. Ham’s F-12 Nutrient Mixture without Hypoxanthine can also be used to culture myeloma cells, hybridomas, primary rat hepatocytes, and prostate epithelial cells.
Recommendation
Add L-glutamine prior to use at a final concentration of 1.0mM.
Supplement 10% FBS because F-12 contains no proteins or growth factors.
